Cancer cell mitochondria are the powerhouse and Achilles heel of tumor growth and metastasis. Cancer cells consume more than 5 times the energy than normal cells. This is contradictory to Otto Warberg's Nobel Prize winning thesis that cancer cells produced only limited amounts of ATP by burning glucose (glycolysis). Apparently, Warberg was measuring the combined energy production of both cancer cells and their surrounding stromal cells. Warberg believed that cancer cells derived their energy from sugar, but it turns out that it is the mitochondria that is making ATP. This effect may also explain why cancer patients lose weight as they get closer to dying. Cancer cells will form when cells lose their ability to burn glucose and oxygen together in the little organelles called mitochondria. When enough of the mitochondria become faulty, a cell will go into a fermentative state or glycolysis to maintain that one billion ATP. This is the central action that causes energy to be produced in biology. The mitochondria are used for energy as it is a more efficient to produce it. One molecule of glucose will produce 38 ATP. In comparison, once glycolysis occurs, a single molecule of glucose will make just two ATP, and this is very inefficient. While in a fermentative state, a cell must obtain a lot more fuel (glucose) to sustain the metabolic requirement that is needed. Once this happens, there will be cancer cells upregulating and growing on their surface in order to have many more insulin receptors which it needs to capture more glucose. Insulin is crucial to opening the door of the cell which allows the glucose inside. When there is too much glucose entering the cells, they can become resistant to insulin which leads to diabetes. This is a defensive measure taken by the body to prevent an excessive amount of glucose from entering the cells. Cristina Muñoz Pinedo. Cell Death Regulation Group IDIBELL (Institut d'Investigació Biomèdica de Bellvitge) Oncogenic transformation promotes metabolic changes which makes tumors "addicted" to certain metabolites. For this reason, inhibition of tumor metabolism is a promising new therapeutic approach. However, little is known about how metabolic stress triggers tumor cell death. Glucose depletion has been shown to kill cells either by necrosis (non-apoptotic, pro-inflamatory cell death) or by the mitochondrial pathway of apoptosis. Our studies indicate that several tumor cell lines of different origins die in a non-apoptotic manner when deprived of glucose. However, we have recently described an atypical apoptotic pathway engaged in cells from solid tumors. Surprisingly, apoptosis induced by glucose deprivation is independent of the Bcl-2-regulated mitochondrial pathway. We will describe this apoptotic pathway mediated by caspase-8, which is the initiator caspase engaged by death receptors of the TNF family. 2-deoxyglucose is a non-metabolizable glucose analog which competes with glucose and has shown anti-tumor effects in animals. Moreover, this compound is been tested in clinical trials. We are currently studying sensitivity of tumor cells, especially sarcoma cell lines, to 2-deoxyglucose. Interestingly, 2-deoxyglucose promoted apoptosis in cell lines in which glucose deprivation promoted necrosis, suggesting different death mechanisms. 2-deoxyglucose activates the mitochondrial apoptotic pathway and regulates several apoptotic proteins in p53-deficient cells. We found death under normoxia to be associated to endoplasmic reticulum stress rather than lack of ATP. We will discuss the signaling pathways involved in responses to nutritional stress and how to improve sensitivity of tumor cells to metabolic targeting.

Cancer breakthrough: CHILLIES can 'self-destruct' cancer cells - scientists discovery. Natural Compound, Capsaicin in HOT SAUCE Can KILL CANCER CELLS. Capsaicin, the active ingredient that gives chillies their bite, surrounds the deadly growths and makes them 'self-destruct', scientists have claimed. The research, from Ruhr University in Bochum, Germany, found the eye-watering component can trigger certain defenses in the cell membranes that surround cancerous cells. "For example, osteosarcoma, colon, and pancreatic cancer cells, while normal cells remain unharmed." According to scientists, the active tongue-tingling ingredient triggers a cell receptor known as TRPV1, which influences the substances cancer cells can feed on. In turn, the cancerous growth fights to battle it and eventually self-destructs. As they all self-destruct, the growth of the cancerous tumour is completely stunted. But before you go stocking up on chilies - there's a catch. Researchers say capsaicin isn't effective if eaten.. Instead it must be formulated into an oral tablet and form of a wider anti-cancer treatment, alongside other drugs that target the cells. Dr Weber explained: "In this study, we aimed to identify the TRP channels in different breast cancer subtypes and to investigate the effect of TRPV1 ligand (capsaicin) on breast cancer progression. "To our knowledge, no studies have yet conducted a large-scale comparative study of the TRP channels expression profiles in breast cancer cell lines. "In all our experiments, a significant reduction in cell proliferation after capsaicin stimulation was observed. "This finding was in accordance with the results of other scientists, who demonstrated a significant decrease in the cell growth rate of MCF-7 breast cancer cells upon capsaicin stimulation." Reportedly, more than 9,000 cancer patients have waited longer than they should since the SNP launched a 'new approach' to beating the disease. Why are immune cells able to kill viruses and bacteria but not cancer cells? Lenneke Cornelissen researches how cancer cells are able to evade the body’s immune attack. With her obtained knowledge, we might be able to cure more cancer patients in the future with immunotherapy. Lenneke Cornelissen is a talented, young researcher on cancer immunology at VU Medical Center in Amsterdam. Our immune cells can attack and kill cancer cells, but unfortunately cancer cells suppress immune responses and thus win the fight. Boosting the patient’s own immune system to fight the tumor is the major goal of cancer immunotherapy. It is very promising and it has cured patients which failed to respond to all standard treatments. Nevertheless, it is not successful in all patients and there is still a lot of work to be done in order to understand how we may improve cancer immunotherapy. Talk Overview: Dr. Fuchs begins her talk with a brief history of stem cells including the discovery in the 1970s that adult skin stem cells could be cultured in vitro indefinitely. This early work provided the foundation for later advances in embryonic stem cell (ESC) culture. ESCs are special because they can generate all the different tissues of the body, thus providing great potential for use in regenerative medicine. The use of ESCs is controversial, however, so scientists have developed ways of generating pluripotent stem cells that do not use embryonic tissue. Fuchs reviews methods such as nuclear transfer and the generation of induced pluripotent stem cells (iPSCs) by reprogramming adult somatic cells. Cells generated by these methods may be used for drug and therapy screening and they may provide treatments for human diseases such as macular degeneration, Parkinson’s and other degenerative diseases. In her second talk, Fuchs focuses primarily on studies of adult skin stem cells. Adult stem cells have the ability to make more stem cells and to generate the cells of a differentiated tissue. Skin stem cells can replenish the epidermis and make hair follicle cells. Skin grown in culture from just a few skin stem cells can be used to treat burn patients or replace damaged corneal epithelium. Stem cells reside in specific niches or microenvironments and signals from the niche determine whether a stem cell is going to be quiescent or make tissue. Fuchs’ lab has studied hair follicle stem cells for many years and has identified the signals that activate hair follicle stem cells as well as the specific set of transcription factors that are upregulated in the activated stem cells. They have shown that many of the genes turned on during stem cell activation are regulated by super enhancers which sense the niche environment. Squamous cell carcinomas (SCCs) are commonly occurring, dangerous cancers that may originate from skin stem cells. By developing methods to identify the stem cells that will lead to cancer, Fuchs’ lab has been able to study how these cells differ from normal skin stem cells. They found that the gene expression profile in normal versus cancer stem cells is very different and is likely the result of differences in the cancer stem cell niche. The tumor microenvironment may include immune cells and greater proximity to TGF-beta secreting blood vessels. Fuchs’ lab showed that cancer stem cells exposed to TGF-beta had increased expression of proteins in the glutathione metabolism pathway; the same pathway which is involved in the breakdown of chemotherapy drugs. Further studies in SCC patients, showed a strong correlation between increased mRNA levels for glutathione pathway proteins and decreased survival. These results suggest that drugs which block TGF-beta activity, in combination with other chemotherapeutics, may be an effective therapy for some SCC. Speaker Biography: Elaine Fuchs is the Rebecca C. Lancefield Professor of Mammalian Cell Biology and Development at The Rockefeller University and an Investigator of the Howard Hughes Medical Institute. Her lab studies the role of skin stem cells in homeostasis and wound repair and how these processes go awry in cancer and inflammatory diseases. Fuchs received her BS in chemistry from the University of Illinois, Champaign-Urbana and her PhD in biochemistry from Princeton. She developed her interest in skin biology as a post-doctoral fellow with Howard Green at the Massachusetts Institute of Technology. Before joining Rockefeller University in 2002, Fuchs was a faculty member at the University of Chicago for twenty years. Fuchs’ many contributions to skin and stem cell biology have been recognized with myriad awards and honors including the National Medal of Science in 2008, the March of Dimes Prize in 2012, and the E.B. Wilson Prize from the American Society of Cell Biology in 2015, to name a few. She is a member of the National Academy of Sciences and the National Academy of Medicine. Dr. Charles Majors recovered from life threatening cancer. Diet was a huge part of that recovery. Here he talks about sugar and what it does to the body and the cells themselves. He also talks about what might be worse than sugar. Transcript Scott: Just how bad is sugar, when it comes to cancer? We know that PET scans can detect it by using sugar, but when people continue to consume sugar, what's going on? Dr. Charles Majors: Well, some people always say 'what's worse, sugar or bad fats'? Bad fats are worse, because it's the bad fats that create more of the inflammation. And then the cell will convert over into only burning sugar. Now, too much sugar can eventually create inflammation and raise the insulin levels. So if we really broke it down and say, 'Ok, we know based upon PET scanning that cancer cells use more sugar than a normal cell'. Otherwise ask your doctor or oncologist don't use... can you not give me sugar or glucose? They're going to be like, well, we couldn't find it. So, that's gone. If your doctor says it doesn't matter what you eat, they're absolutely wrong. So, number two is, we know that a normal, healthy cell has mitochondria, healthy mitochondria. The mitochondria are what bring in the fuel and create this ATP. ATP is your energy source. So, think of the mitochondria as your engine in your car. The engine has to be working perfectly. And the engine, on normal, healthy cells, bring in oxygen and they bring in glucose. For every one molecule of glucose though... because it is such an efficient engine think of it as a smart car, right? These healthy cells have a smart mitochondria, perfect mitochondria, and they are able to bring in, for every one molecule of glucose, they are able to create 36 ATP. Just amazingly fuel efficient. And we look at the difference in a cancer cell. And because their mitochondria is damaged, they no longer can bring in oxygen for their fuel source. So they are horrible, it's the most... it would be like having a Humvee, you know what I mean, for inside of there. It's getting two miles to the gallon here. For every one molecule of glucose, it can only make about two ATP. So it's very inefficient. So in order for these cells to survive, they have to pop up, you know, ten to twelve to 18 times more insulin receptor sites... and so to bring in more sugar to those cells. So we know just based on that, that these cells, normal cells are just more efficient, they don't need sugar. These cells need sugar for their energy. So now we say, well what about fruit. This is what I get all the time. I haven't had fruit... I mean actually, now that its been almost five years, I'm able to put a little bit of fruit back in, but I follow ketogenic diet, based upon the metabolism of cancer. And so I don't bring in much fruit. Now does fruit feed cancer? No. Fruit, the fruit sugars spins left, where the regular processed sugars spin right, so cancer cells don't use that. But what fruit does do is, and this is the argument you get... you need fruits in ahh.... fruit will raise insulin levels. You ask any diabetic, they can't eat a lot of fruit. So if you raise an insulin level, insulin will create more inflammation. Insulin will take me out of ketosis. I need my body the most efficient, fuel burning. And I want it burning keytones. And so whenever my insulin levels go up, my body goes out of ketosis. My liver's producing keytones, I've been doing it for five years. I've been in ketosis, and still alive, doing great. Your normal, healthy cells will convert over to using keytone for their fuel, and you know cancer cells can't use them. So for me, it's another way of starving this cancer, because I don't give it anything. There's no glucose coming in. I have to watch even how much protein I bring in because glutamate will feed into those cancer cells. You have to watch how much sugars, how much proteins and I increased it all to higher fats. I watch all, I know no high glycemic vegetables at all.
